【 Principle 】
Peptone and beef extract provide nitrogen sources and growth factors such as amino acids, peptides, and vitamins, meeting the basic growth and metabolic needs of Staphylococcus aureus; Mannitol is a fermentable carbohydrate substrate used for biochemical identification;
High concentration sodium chloride forms a strong and high osmotic pressure environment, inhibiting the vast majority of salt intolerant bacteria such as coliforms, streptococci, fungi, etc. Bromothymol purple serves as an acid-base indicator, and Staphylococcus aureus can
ferment mannitol to produce organic acids. As the pH decreases, the indicator changes from purple to yellow; Coagulase negative staphylococci such as Staphylococcus epidermidis cannot ferment mannitol, produce no acid, and maintain the purple color of the culture medium
unchanged.
Main components
Composed of peptone, sodium chloride, beef extract, mannitol, bromothymol violet, and distilled water.
Storage conditions and expiration date
Storage conditions: This product should be stored in a dark environment at 2-8 ℃.
Validity period: The unopened original packaging has a validity period of 6 months.
This product is a disposable product.
Selective isolation and screening of Staphylococcus aureus: isolation and identification of pathogenic Staphylococcus in clinical pus, wound secretions, sputum, and blood samples.
Distinguishing pathogenicity from normal skin staphylococci: relying on mannitol fermentation to change color, quickly distinguish Staphylococcus aureus from coagulase negative staphylococci such as epidermal and saprophytic staphylococci.
Hospital infection monitoring: Sampling of medical staff's hands, ward environment, and medical device surfaces to screen for drug-resistant Staphylococcus aureus such as MRSA.
Microbial testing for food hygiene: detection of Staphylococcus aureus contamination in cooked food, meat products, dairy products, and pastries.
Auxiliary diagnosis of urinary and skin infections: Selective culture of Staphylococcus aureus in urine and skin swab specimens to exclude interference from other bacteria.
Staphylococcus aureus
Gram positive cocci, arranged in clusters of grapes, without spores or flagella, some with capsules; It can withstand a high osmotic pressure of 7.5% sodium chloride, which is its key biological characteristic; Can ferment mannitol to produce acid; Phenol red indicator turns yellow when exposed to acid, and the culture medium/liquid turns yellow; Facultative anaerobic, low nutritional requirements, common pathogenic bacteria in clinical wounds, blood, and food.
Staphylococcus epidermidis, Staphylococcus saprophyticus
Gram positive Staphylococcus aureus, without spores; Can also tolerate high salt environments and grow on culture media; Cannot ferment mannitol, does not produce acid, and maintains the original culture medium; Mostly composed of normal skin microbiota, Staphylococcus epidermidis is prone to causing infections in catheters and implants; Staphylococcus saprophyticus often causes urinary tract infections.
