Casein peptone, beef extract, and yeast extract provide amino acids, peptides, B vitamins, and growth factors; Glucose provides a carbon source; Sodium chloride maintains osmotic pressure; L-Cysteine and sodium thioglycolate are reducing systems that consume free oxygen in the system, reduce the redox potential, and create an anaerobic living environment for anaerobic bacteria; Simultaneously protecting anaerobic bacteria from oxidative damage caused by oxygen; Low concentration agar increases the viscosity of the culture medium, slows down the downward permeation of oxygen in the air, and naturally forms an oxygen gradient of aerobic in the upper layer and anaerobic in the lower layer. Aerobic bacteria grow in the upper layer, while anaerobic bacteria grow in the lower layer; Methylene blue is an oxidation-reduction indicator.
