[Testing Principle]

Casein peptone is rich in free amino acids and short peptides, providing balanced and stable nutrition that can supply essential nutrients for the growth of most non-demanding aerobic and facultative anaerobic bacteria. Beef extract powder supplements carbon sources, inorganic salts, and vitamins, enhancing the nutritional system. Soluble starch adsorbs heavy metal ions and trace toxic impurities in the culture medium, eliminating interference from impurities with antibiotics and bacteria, buffering harmful substances produced by bacterial metabolism, stabilizing bacterial growth, and ensuring uniform colony formation. Agar serves as the gelling agent.

Introduction to the Relevant Microbiological Characteristics

Escherichia coli

G⁻ facultative anaerobic bacillus, an opportunistic pathogen in the urinary and intestinal tracts; lactose-fermenting, with mostly motile; colonies of moderate size, gray-white, smooth, moist, and convex with a translucent appearance.

Klebsiella genus

G⁻ facultative anaerobic bacillus, non-flagellated, with a capsule; common pathogenic bacteria found in sputum and wound exudates; colonies are large, grayish-white, highly viscous, and exhibit obvious stringing with a loop inoculation.

Staphylococcus aureus

*Streptococcus pyogenes*, facultative anaerobe, plasma coagulase-positive, causing skin and wound infections; colonies of medium size, golden/yellowish-white, opaque, smooth and raised.

Salmonella, Shigella

G⁻ facultative anaerobic pathogenic bacteria, non-capsulated (except for some Salmonella), causing intestinal infections; colonies are small to medium in size, smooth, translucent, and gray-white.
