Product Usage:For the recovery of Bacillus stearothermophilus spores, it is commonly used in steam sterilization bioindicator identification tests during disinfection efficacy testing.
Product Overview

【 Principle of Inspection 】

Protein peptone and beef extract provide abundant nitrogen sources and growth factors, promote the repair and germination of heat damaged spores, and avoid false negatives; Glucose is a fermentable carbon source, exclusively used for the metabolism and acid production of thermophilic fat bacteria, while rapidly supplying energy; Soluble starch serves as a buffering and protective agent, adsorbing trace heavy metals and toxic impurities in the culture medium, reducing toxicity inhibition on damaged spores, and stabilizing the pH environment of the culture medium; Agar is a coagulant.

Main components

Composed of peptone, beef extract, soluble starch, glucose, and agar.

Storage conditions and expiration date

Storage conditions: This product should be stored in a dark environment at 2-8 ℃.

Validity period: The unopened original packaging has a validity period of 6 months.

Clinical application scenarios

Sterilization effect verification: Cultivate thermophilic Bacillus spores to determine if sterilization is qualified.

Daily sterilization monitoring: Weekly/batch monitoring in the operating room and supply room to determine if sterilization is ineffective.

Medical device sterilization validation: testing the reliability of consumables and device sterilization processes.

Equipment troubleshooting: reproduce spore growth and assist in analyzing sterilization equipment issues.

Indicator quality inspection: detect spore vitality and heat resistance, evaluate whether the indicator meets the standard.

Sterilization confirmation for pharmaceutical factories: Suitable for high-temperature spores to avoid false negatives and ensure sterile production.

Introduction to the Relevant Microbiological Characteristics

Thermophilic fat loving Bacillus subtilis

Biological characteristics: Gram positive Bacillus, aerobic, producing endophytic spores; Thermophilic bacteria, with an optimal cultivation temperature of 55-60 ℃; Spores have extremely strong heat resistance and are standard biological indicator bacteria for pressure steam sterilization, withstanding saturated steam at 121 ℃; Spores damaged during incomplete sterilization can be repaired, germinated, and reproduced in this culture medium; Decompose sugars to produce acid, with no strong pathogenicity, is an environmental saprophytic bacterium, and does not cause human infection.

Colony characteristics: Medium size, round, flat or slightly raised, with neat and smooth edges; Milky white, opaque; Yellow color circles may appear around the culture medium of the colony (the strain produces acid, and the culture medium contains a pH indicator); Growing rapidly at 56 ℃, clear colonies can be seen with the naked eye within 24 hours; Almost no growth occurs at room temperature of 37 ℃.
