【 Principle 】

Tryptone and soy peptone provide the necessary carbon, nitrogen, vitamins, and growth factors for bacterial growth; Sodium chloride maintains a balanced osmotic pressure; Agar is a coagulant for culture media.

Clinical application scenarios

Colony count detection: a national standard basic plate for counting the total number of aerobic/facultative anaerobic bacteria in samples such as food, water, and equipment.

Strain recovery and purification: Recovery, purification, and passage preservation of standard strains and pathogenic bacteria for subsequent identification.

Purification of bacterial enhancement solution: Selective bacterial enhancement solution turbidity tube is first inoculated with TSA to obtain pure bacterial colonies, and then transferred to identification plates.

Disinfection effectiveness verification: Recovery of live bacteria during sterile inspection and sterilization testing to evaluate disinfection and sterilization capabilities.

Introduction to the Relevant Microbiological Characteristics

Staphylococcus aureus

Biological characteristics: Staphylococcus aureus, facultative anaerobic, salt tolerant, common in skin/food, opportunistic pathogen; TSA colony: round convex, smooth, opaque, golden/milky white in color, medium in size.

Listeria monocytogenes (Listeria monocytogenes)

Biological characteristics: Bacillus subtilis, cold loving, facultative anaerobic, foodborne pathogenic bacteria; TSA colony: small, gray white, semi transparent, smooth edges, can grow slowly at low temperatures of 4 ℃.

Bacillus cereus

Biological characteristics: Bacillus subtilis, producing endophytic spores, aerobic, food contaminating bacteria; TSA colony: gray white, large and flat, ground glass like, with irregular edges.

Escherichia coli

Biological characteristics: G ⁻ short rod-shaped bacteria, facultative anaerobic, lactose fermentation, fecal indicator bacteria; TSA colony: gray white, smooth and moist, medium size, no special pigments.

Salmonella (Salmonella enterica, Salmonella enterica)

Biological characteristics: Gram positive flagellar bacteria, foodborne pathogens, do not ferment lactose; TSA colony: colorless, transparent, smooth, round, gray white, without black sediment.

Klebsiella genus

Biological characteristics: G ⁻ crude short rod-shaped bacteria, with capsules and mucous type; TSA colony: large, sticky and protruding, with obvious wire drawing, gray white in color.
