【 Principle of Inspection 】
Tryptone and bovine heart extract powder serve as the basic nutrients in the culture medium, providing the necessary carbon and nitrogen sources, vitamins, and other growth factors for bacterial growth; Sodium dihydrogen phosphate as buffer: glucose as fermentable carbon source; Sodium chloride maintains the osmotic pressure required for normal growth of bacterial cells.
Main components
Composed of cow heart extract powder, tryptone, sodium chloride, glucose, disodium hydrogen phosphate, and distilled water.

Isolation and cultivation of harsh bacteria: primary isolation of pathogenic bacteria with high nutritional requirements, such as Streptococcus pneumoniae, Streptococcus pneumoniae, Listeria monocytogenes, Neisseria gonorrhoeae, and Corynebacterium;
Anaerobic bacterial culture: in conjunction with anaerobic environment, clinical anaerobic pathogenic bacteria are isolated and purified;
Drug susceptibility test: Pure cultivation of harsh and anaerobic bacteria before drug susceptibility testing;
Strain preservation: valuable standard strains and clinical isolates are passaged and preserved;
Blood and sterile body fluid specimens for bacterial enrichment and transfer: Blood culture positive bottles are transferred to plates for pure bacterial separation.
Staphylococcus aureus
G ⁺ cocci, with a diameter of about 0.8-1.0 μ m, arranged in grape clusters, without spores, flagella, or capsule formation; Aerobic or facultative anaerobic, suitable for growth temperature of 37 ℃, suitable for pH 7.4; Strong salt tolerance; Catalase positive; Decompose glucose, maltose, lactose, sucrose, produce acid but not gas; Methyl red reaction positive, VP reaction weakly positive. After culturing at 35-37 ℃ for 18-24 hours, the broth showed uniform cloudy growth (turbidity 2) and evenly distributed bacterial cells; BHI broth does not contain blood components, therefore it does not exhibit hemolysis.
Escherichia coli
G ⁻ short rod-shaped bacteria, facultative anaerobic, ferment lactose to produce acid and gas; After 8-24 hours of cultivation, the broth becomes uniformly turbid.
Listeria monocytogenes
G ⁺ short rod-shaped bacteria, cold loving facultative anaerobic, foodborne pathogenic bacteria; BHI cultivation, uniform to moderate turbidity, without obvious large sediment.
Streptococcus pneumoniae
G ⁺ - Streptococcus, facultative anaerobic, nutrient demanding, requires abundant growth factors, respiratory pathogenic bacteria; The BHI culture liquid is slightly and uniformly turbid, with a small amount of fine white sediment at the bottom of the tube and no gas production.
