Purification of suspected Vibrio colonies: Select suspicious colonies from TCBS, color plates, or bacterial enhancement solution and transfer them to obtain pure colonies, providing pure culture for biochemical and serum identification.
Purification passage and short-term storage: As a high salt nutrient plate without strong inhibitors, it is used for Vibrio passage recovery and short-term laboratory storage.
Preparation of pure bacteria before drug sensitivity test: After purification, Vibrio is revived on this plate to prepare a standardized bacterial suspension, ensuring accurate drug sensitivity results.
Isolation of Vibrio from Aseptic Samples: Direct inoculation of pus, ascites, blood, etc. from wounds caused by seawater puncture is beneficial for the growth of Vibrio vulnificus and Vibrio alginolyticus.
Assisted identification of halophilic characteristics: Combined with regular TSA control: only 3% salt plate growth, no salt growth, preliminary differentiation between Vibrio and common Escherichia coli.

Vibrio parahaemolyticus
Biological characteristics: Gram negative arc-shaped, strictly halophilic, no growth without salt; Exists in marine products, the pathogenic strain Kanagawa is positive, causing acute gastroenteritis.
Cultivation characteristics: Medium to large, gray white opaque colonies, with a sticky texture; Some strains exhibit beta hemolytic rings; The growth rate is slightly faster than that of Vibrio cholerae.
Vibrio cholerae
Biological characteristics: Gram negative arc-shaped, single ended single flagella, strong motility; 0% to 3% NaCl can grow and is facultative anaerobic; Fermented sucrose produces enterotoxins, causing severe watery diarrhea.
Cultivation characteristics: Medium sized, milky white, smooth and moist raised colonies at 37 ℃ for 18-24 hours, semi transparent, with neat edges, no pigmentation, and no hemolysis (slight beta hemolysis in a few strains).
Vibrio vulnificus
Biological characteristics: Halophilic Vibrio, distributed in seawater; Wound contact or ingestion infection can easily cause soft tissue necrosis, sepsis, and high mortality rate.
Cultivation characteristics: Small to medium gray white colonies, some strains show weak green pigments, and a few have hemolysis.
