【 Principle 】

Protein peptone, pancreatic peptone, and yeast extract provide carbon, ammonia, vitamins, and growth factors; Sodium amide maintains a balanced osmotic pressure; Sodium pyruvate, sodium metabisulfite, and ferrous sulfate are beneficial for the growth of Campylobacter. Cefoperazone can inhibit Gram negative bacteria and some Gram positive bacteria, amphotericin B can inhibit fungal growth, and rifampicin can inhibit non curved bacteria and other anaerobic bacteria; Agar is a coagulant for culture media.

Main components

It is composed of peptone, tryptone, yeast extract, sodium chloride, sodium pyruvate, sodium metabisulfite, ferrous sulfate, cefoperazone, polymyxin B, rifampicin, agar, ethanol, sterile water, sterile defatted sheep blood, and distilled water.

Clinical application scenarios

Primary isolation of Campylobacter from diarrhea patients: targeting the feces of patients with abdominal pain, watery/bloody stools, and fever, with 42 ℃ microaerophilic culture, it is a classic blood agar plate.

Isolation of low load damaged specimens: Sheep blood provides abundant nutrients, buffers oxygen damage, and increases the detection rate of low bacterial count and damaged bacterial cells.

Parallel screening with mCCD: using bloodless CCD in combination with blood containing Skirrow, balancing cost and sensitivity, and reducing missed detections.

Source tracing of foodborne poisoning: cultivation of livestock and poultry meat, drinking water, and vomit to detect pathogenic curved bacteria for epidemiological tracing.

Cultivation of zoonotic specimens: screening of animal manure and slaughter environment samples to assess the risk of infection at the source of food ingredients.

Purification of bacterial colonies for identification of drug sensitivity: Select typical single bacterial colonies for purification, and use them for oxidase, uric acid hydrolysis, serum typing, and drug sensitivity testing.

Introduction to the Relevant Microbiological Characteristics

Campylobacter jejuni, Campylobacter coli

Biological characteristics: Gram negative, arc-shaped, S-shaped, seagull spreading filamentous bacteria, single/double ended flagella, shuttle like high-speed movement; Strictly microaerophilic (5% O ₂, 10% CO ₂, 85% N ₂), most suitable for 42 ℃, no growth at 25 ℃; Positive for oxidase and peroxidase; Campylobacter jejuni is positive for uric acid hydrolysis, while Campylobacter coli is negative; Not fermenting sugars, not producing H ₂ S; mainly carried by poultry, with foodborne acute hemorrhagic enteritis, complications can induce Guillain Barr é syndrome.

Skirrow blood agar culture characteristics: microaerophilic at 42 ℃ for 24-48 hours; colonies are small, flat, gray white semi transparent, with loose and irregular edges, often in the form of spreading mist or water droplet like films; Most strains have no obvious hemolysis, while a few strains have mild alpha hemolysis; Unique damp and moldy odor; Pick up bacterial colonies and smear them, and observe the characteristic curved S-shaped bacteria. The suspension is highly dynamic.
