[Sample Requirements]
Samples such as sputum, blood, urine, cerebrospinal fluid, and wound secretions from the human body.
Primary isolation of clinical specimens: suitable for blood, pus, sputum, cerebrospinal fluid and other specimens, and can be used to culture common bacteria and harsh culture bacteria.
Observation of Hemolysis Characteristics: Rapid identification of Group A Streptococcus, Streptococcus pneumoniae, etc. through alpha, beta, and gamma hemolysis to assist in initial screening of bacterial strains.
Purification of suspicious colonies: Select suspicious colonies from the selective plate and transfer them to obtain pure colonies for subsequent identification.

Streptococcus pyogenes

Cultivation characteristics: At 35 ℃ for 24 hours, small gray white raised colonies with clear and transparent beta hemolytic rings around the periphery of the colonies; Enzyme negative.
Streptococcus pneumoniae
Biological characteristics: Gram positive diplococcus, arranged in pairs in a pointed shape, with capsules; Alpha hemolysis can cause pneumonia, meningitis, and otitis media.
Cultivation characteristics: Flat, semi transparent needle like colonies surrounded by grass green alpha hemolytic rings; Bile lysate is positive.
Listeria
Biological characteristics: Gram positive Bacillus subtilis, motile at 25 ℃, a zoonotic pathogen that can cause sepsis and meningitis.
Cultivation characteristics: small blue gray colonies, narrow beta hemolytic ring; 4 ℃ can increase bacterial growth by cold treatment, and the contact enzyme is positive.
Staphylococcus (Staphylococcus aureus, coagulase negative Staphylococcus)
Biological characteristics: Gram positive Staphylococcus aureus, facultative anaerobic; Staphylococcus aureus produces various toxins, causing skin infections and bloodstream infections.
Cultivation characteristics: large golden/milky white colonies; Staphylococcus aureus shows a wide beta hemolytic ring, while Staphylococcus epidermidis is mostly gamma hemolytic (without hemolysis) and positive for lysozyme.
Haemophilus influenzae (Haemophilus influenzae, etc.)
Biological characteristics: Gram negative short ball bacillus, strictly requires X and V factors, belongs to the category of harsh bacteria, causing respiratory and meningeal infections.
Cultivation characteristics: transparent colonies resembling small dewdrops, without hemolysis; It relies on heme and coenzyme in sheep blood to grow.
