Bacterial count of specimens: determination of the total number of bacteria in sewage, drinking water, medical equipment, environmental surfaces, and disinfection effectiveness monitoring;

Staphylococcus aureus: Gram positive cocci, typically arranged in "grape clusters", without spores or flagella, and some strains can form capsules; Rapidly growing on nutrient agar, forming round, raised, smooth and moist colonies on the surface; The diameter is about 2-3mm, with a typical golden yellow color. Transparent hemolytic rings may appear around the colony, and there is no special odor. It is enzyme positive and can decompose glucose, maltose, and other substances to produce acid and gas, but does not decompose lactose; It can produce plasma coagulase, has strong tolerance to dryness and salt, and can still grow in an environment of 10% to 15% sodium chloride.
Escherichia coli: Gram negative short rod-shaped bacteria without spores and with multiple flagellar pili. It is facultative anaerobic and grows into milky white smooth medium-sized colonies on nutrient agar. It can ferment glucose and lactose to produce acid and gas, and has an IMViC of++- - -. It contains O/H/K antigens, is not resistant to high fever, and is easily inactivated by conventional disinfectants. Most of them are normal gut microbiota, and pathogenic subtypes can cause intestinal and extra intestinal infections.
