【Principle】

Tryptone provides nitrogen sources, vitamins, and amino acids; soybean peptone supplements growth factors and carbohydrates lacking in tryptone; glucose serves as the carbon source; sodium chloride maintains balanced osmotic pressure; potassium hydrogen phosphate acts as a buffer; and agar functions as the solidifying agent in the medium.

【Main Ingredients】

Composed of pancreatic trypticase, soybean trypticase, sodium chloride, potassium hydrogen phosphate, glucose, agar, and distilled water.

Storage conditions: This product should be stored in a light-protected environment at 2-8°C.

Clinical application scenarios

Primary isolation of clinical specimens: Suitable for culturing purulent exudate, sputum, wound secretions, etc., enabling simultaneous cultivation of both common bacteria and mildly fastidious organisms.

Hemolysis-assisted preliminary screening: Rapid detection of characteristic strains such as Streptococcus pyogenes, Listeria, and Staphylococcus aureus through α, β, and γ hemolysis.

Selective plate colony purification: Suspected colonies were picked from plates such as MYP, PALCAM, and mCCD and subcultured to obtain pure colonies for subsequent identification.

Listeria monocytogenes reexamination: Observe the characteristic narrow β-hemolytic ring as the key identification test on the agar plate.

Revival and activation of damaged strains: Glucose and sheep blood synergistically repair bacteria damaged by refrigeration and disinfection, enhancing the detection sensitivity of low-load specimens.

Standardization of pre-antimicrobial susceptibility strains: Purified strains are subcultured and revived for use in disk diffusion or broth dilution antimicrobial susceptibility testing to ensure reliable results.


Introduction to the Relevant Microbiological Characteristics

Listeria monocytogenes

Biology: Gram-positive short bacillus, facultative anaerobe; exhibits motility with peritrichous flagella at 25°C but no motility at 37°C; catalase-positive, capable of producing narrow β-hemolysis; psychrophilic, capable of proliferation at 0–4°C; a high-risk foodborne pathogen, often causing invasive infections in pregnant women and newborns.

TSSB Culture Characteristics (30~37°C, 24h)

Fine, blue-gray, translucent colonies with a narrow, distinct beta-hemolysis zone around the periphery; colonies are moist and show slight spreading.

Bacillus cereus

Biology: Gram-positive, large, filamentous bacillus, spore-forming, with non-swollen spores; lecithinase-positive, does not ferment mannitol, facultative anaerobic; causes vomiting and diarrhea-type food poisoning in contaminated rice and flour products.

TSSB cultivation characteristics: Gray-white, flat, rough large colonies with irregular edges. Most strains show no hemolysis (γ-hemolysis), while some exhibit weak β-hemolysis.

Streptococcus pyogenes

Biological characteristics: Gram-positive cocci arranged in chains; facultative anaerobe with high nutritional requirements; produces streptolysin O and streptolysin S; Pathogenicity: causes pharyngitis, scarlet fever, impetigo, rheumatic fever, etc.

TSSB culture characteristics: colonies are gray-white, smooth, with diameters of 0.5-1.5 mm; complete lysis of red blood cells around the colonies forms a clear transparent zone; β-hemolysis.
