Tryptone provides carbon and nitrogen sources; Lactose is a fermentable sugar in the Escherichia coli community; Potassium dihydrogen phosphate is a buffering agent; Agar is a coagulant for culture media; Yihong and Meilan are antibacterial agents and pH indicators that can inhibit Gram positive bacteria, produce precipitates under acidic conditions, and form purple black colonies or colorless transparent colonies with black centers on the periphery.

Intestinal infection screening: isolation and culture of fecal samples to identify pathogenic bacteria such as Escherichia coli, Salmonella, and Shigella that cause diarrhea.
Urinary tract infection testing: Urine culture, screening for common pathogenic bacteria such as Escherichia coli and Klebsiella in the urinary system.

Escherichia coli
Biological characteristics: G ⁻ short rod-shaped bacteria, with flagella around the body, mostly with pili, no spores, and some strains have capsules; Facultative anaerobic; Biochemistry: Fermentation of glucose, lactose to produce acid and gas, IMViC:++--; Urinary tract, intestinal, abdominal, and bloodstream infections can cause diarrhea and acute enteritis in the large intestine.
Colony: purple black with typical green metallic luster, medium size.
klebsiella pneumoniae
Biological characteristics: G ⁻ crude short rod-shaped bacteria, thickened capsule, flagellar and spore free, facultative anaerobic; Fermented lactose produces acid, with mucous colonies; IMViC:--++; Hospital acquired pneumonia, urinary tract infection, biliary tract infection.
Bacterial colony: Large, viscous and brushed, brown black/black brown lactose fermenting bacterial colony Gas producing Escherichia coli
Biological characteristics: flagella, no spores or capsules, facultative anaerobic; Lactose+glucose gas production; IMViC:--++; Conditional pathogenic bacteria, postoperative infections, and urinary tract infections.
Colony: Large, dark black, without obvious metallic luster.
Shigella genus (Shigella dysentery, Shigella flexneri, Shigella bovis, Shigella sonnei)
Biological characteristics: G ⁻ short rod-shaped bacteria, without flagella or spores, mostly without capsules, facultative anaerobic; Only ferment glucose, not lactose; No power; Bacterial dysentery invades the colonic mucosa.
Colony: Small, colorless, transparent, and smooth colony; Salmonella genus (Salmonella typhimurium, Salmonella typhimurium)
Biological characteristics: Bacillus subtilis, with flagella, motile, and no spores; Most do not ferment lactose, glucose produces acid and most produce gas (Salmonella typhi only produces acid and not gas); Febrile fever, paratyphoid fever, food poisoning, sepsis.
Colony: colorless, semi transparent, medium-sized.
