【 Principle 】
Acetamide provides the carbon and nitrogen sources required for bacterial growth; Sodium amide maintains osmotic pressure balance in the culture medium system; Phosphates as buffering agents; Magnesium ions as auxiliary factors; Phenol red serves as an acid-base indicator; Agar as a coagulant; Bacteria using acetamide can grow on this medium and produce alkali to turn the indicator color red.
Main components
Composed of acetamide, sodium chloride, anhydrous potassium dihydrogen phosphate, anhydrous dipotassium dihydrogen phosphate, magnesium sulfate, phenol red, agar, and distilled water.

Microbial limit testing for drugs - according to the Chinese Pharmacopoeia, it is an important step in drug quality control for the selective isolation and confirmation of Pseudomonas aeruginosa in drugs.
Cosmetics hygiene inspection: used for selective isolation and cultivation of Pseudomonas aeruginosa in cosmetics samples to evaluate product hygiene and safety.
Monitoring of packaged drinking water quality: commonly used for detecting Pseudomonas aeruginosa in packaged drinking water, combined with methods such as CN agar can reduce false negative results and improve detection reliability.
Rapid testing of hospital preparations: The use of acetamide slant culture medium method can quickly detect Pseudomonas aeruginosa in external drugs, eye drops and other hospital preparations, with simple operation.
Clinical microbiological identification: used in clinical laboratories to distinguish Pseudomonas aeruginosa from other non fermenting gram-negative bacteria.
Screening of cystic fibrosis patients: In specimens of cystic fibrosis patients, using acetamide broth for enrichment and then transferring to selective plates can improve the detection rate of Pseudomonas aeruginosa.
Pseudomonas aeruginosa
Biological characteristics: G ⁻ straight or curved rod-shaped bacteria, single ended flagella, motile, non spore forming, obligate aerobic; Producing acetamide enzyme, hydrolyzing acetamide to produce acetic acid and ammonia, and increasing pH. Growing solely on acetamide as the sole nitrogen source, most other bacteria cannot utilize it; Widely present in humid environments, water sources, cosmetics, and opportunistic pathogens.
Cultivation characteristics of acetamide liquid medium: The culture conditions are 30-35 ℃ and 18-24h, and the color can be significantly changed; Decompose acetylamine to produce ammonia, the culture medium turns rose red, uniformly turbid, and the bottom of the tube is gray white precipitate; Negative for miscellaneous bacteria, unable to decompose acetamide, the culture medium remains light yellow, clear or weakly growing, and does not turn red.
