【 Principle of Inspection 】
Peptone provides amino acids, peptides, nitrogen sources, and trace growth factors to meet the basic growth and metabolic needs of Pseudomonas aeruginosa; Magnesium chloride and potassium sulfate provide magnesium ions, potassium ions, and sulfate ions, which are essential cofactors for the key enzyme system of Pseudomonas aeruginosa to synthesize pyocyanin; Inducing Pseudomonas aeruginosa to produce pigments and inhibiting the growth of miscellaneous bacteria using glycerol as a specific carbon source; Agar is a coagulant.
Main components
Composed of peptone, magnesium chloride, potassium sulfate, glycerol, and agar.
Storage conditions and expiration date
Storage conditions: This product should be stored in a dark environment at 2-8 ℃.
Validity period: The unopened original packaging has a validity period of 3 months.
This product is a disposable product.
Copper green confirmation identification: Using pyocyanin pigment to distinguish copper green from other non fermenting bacteria.
Clinical infection tracing: Detecting sputum, pus, secretions and other specimens to assist in determining the type of copper green infection.
Microbial testing of drugs: Confirm the presence of copper green positive strains in external drugs, water solutions, etc. according to pharmacopoeia standards.
Monitoring of nosocomial infection environment: Screening for bacterial colonies in ICU, humidification bottles, and other environments, and identifying sources of copper green contamination through staining.
Toxicity assessment of bacterial strains: Distinguish between highly pigmented strains and weakly pigmented strains to determine the risk of disease.
Quality Control and Validation: Used for passaging of copper green standard strains and quality control of culture medium performance.
Pseudomonas aeruginosa
Biological characteristics: Gram negative obligate aerobic bacteria, with a single ended flagellum and active movement; Due to the widespread colonization of opportunistic pathogens in humid environments, they can cause infections in wounds, respiratory tract, urinary tract, burn wounds, and ear infections; Unique metabolic pathway, utilizing glycerol to synthesize pyocyanin and fluorescein; Magnesium sulfate and potassium sulfate are essential ions for the synthesis of pigments; Tolerant to low nutrient environments, most miscellaneous bacteria cannot utilize this culture medium system for large-scale proliferation.
Colony characteristics : 24-48 hours, cultured at 35 ℃,The colony is flat, large, with irregular edges and a moist surface; Blue green/yellow green water-soluble pyocyanin diffuses from the agar around the colony, and the entire plate substrate is stained green; Typical characteristics: ginger like aromatic odor; Under ultraviolet radiation, fluorescein producing strains exhibit yellow green fluorescence; Some weak strains that do not produce pyocyanin only produce light yellow fluorescein, and there is no obvious blue-green color on the agar.
