【 Principle 】

Gelatin trypsin hydrolysate provides nitrogen source, vitamins, and growth factors; Magnesium chloride and potassium sulfate stimulate the production of pyocyanin; Hexadecyltrimethylammonium bromide, as a quaternary ammonium cationic surfactant, changes the permeability of bacteria, causing autolysis or protein denaturation and precipitation, leading to bacterial death. It has strong antibacterial effects, and Pseudomonas aeruginosa has a certain tolerance to hexadecyltrimethylammonium bromide; Agar is a coagulant for culture media.

Pseudomonas aeruginosa

Biological characteristics: Gram negative straight or curved rod-shaped bacteria, single ended single flagella, active motility, aerobic bacteria; Highly tolerant to cetyltrimethylammonium bromide (cationic detergent and antibacterial agent), the vast majority of Gram positive bacteria and other Gram negative miscellaneous bacteria will be inhibited by this reagent; It can produce water-soluble pyocyanin (pyocyanin), positive for oxidase, and can utilize citrate to liquefy gelatin.

Cultivation characteristics (35 ℃, 18-24h): The colonies are round, flat, moist, and have neat edges. Yellow green/blue-green water-soluble pigments diffuse from the culture medium around the colonies; Pseudomonas aeruginosa is soluble in water, and the base color of the tablet is dyed green, with extremely high recognition; Escherichia coli, Staphylococcus, Bacillus, and other bacteria cannot grow, and the background of the plate is clean without interference from other bacteria.
