Peptone and beef extract provide the carbon source, nitrogen source, vitamins, and growth factors required for bacterial growth; Glucose can serve as the energy required for bacterial growth and metabolism; Sodium dihydrogen phosphate plays a role in buffering the pH value of the culture; Sodium sulfite and brilliant green can inhibit the growth of Escherichia coli, Proteus mirabilis, and Gram positive bacteria; Salmonella typhi and other Salmonella bacteria can use glucose to reduce sulfites to sulfides and react with ferrous sulfate to form colonies that appear black. In addition, they can also reduce bismuth ions to metallic bismuth, which gives the colonies a metallic luster and separates Salmonella bacteria.

BS agar colony characteristics: large black or gray black colonies with obvious metallic luster; The culture medium around the colony turns black, with brownish black diffusion and precipitation rings.
Salmonella Enteritidis
Biological characteristics: Gram negative bacteria with flagella; Lactose negative, producing H ₂ S, poultry eggs mainly contaminate serotypes, and are prone to group food poisoning.
BS agar colony characteristics: black circular raised colonies, bright metallic luster, and large areas of surrounding culture medium turning black.
Salmonella paratyphoid B
Biological characteristics: Gram negative bacteria can invade the bloodstream and cause typhoid fever; H ₂ S positive, does not use lactose, urease negative.
BS agar colony characteristics: gray black medium colonies, slightly weak metallic luster, and mild blackening around the periphery.
