【 Principle 】

Peptone and beef extract powder provide sources of ammonia, vitamins, and growth factors; D-mannitol is a fermentable carbon source; Sodium chloride maintains a balanced osmotic pressure; Agar is a coagulant for culture media; Phenol red is a pH indicator; Egg yolk contains phospholipids, and Bacillus cereus produces lecithin enzyme, which forms a precipitation ring around the colony; Does not ferment D-mannitol to produce acid, causing the colony to appear red; Polymyxin B can inhibit the growth of miscellaneous bacteria.

Main components

Composed of peptone, beef extract powder, D-mannitol, sodium chloride, phenol red, polymyxin B, 50% yolk solution, and distilled water.

Clinical application scenarios

Food environment wax sample counting: National standard special flat plate, used for qualitative and quantitative detection of food and processing countertop smear samples such as rice flour and dairy products.

Traceability of food poisoning: Separate and determine whether the aggregated poisoning samples (vomit, leftover food) from vomiting and diarrhea are wax like contamination.

Clinical specimen assisted screening: Separate wax samples from diarrhea feces and contaminated wound secretions to distinguish other intestinal pathogenic bacteria.

Introduction to the Relevant Microbiological Characteristics

Bacillus cereus

Biological characteristics: Gram positive coarse rod-shaped bacteria, with flat ends and a chain like arrangement, can produce elliptical spores, which do not cause the bacterial cells to swell; Phosphatidylase positive, non fermenting mannitol; Enzyme positive, facultative anaerobic; Pathogenic characteristics: Foodborne pathogens, divided into vomiting type and diarrhea type, can cause collective food poisoning by contaminating rice, noodles, and cooked food; Spores are resistant to high temperatures and storage, and can easily reproduce in large quantities when cooked at room temperature.

MYP agar culture characteristics: 30 ℃, 24-48h; colonies pink/light red large colonies (not fermenting mannitol, medium maintained neutral alkaline, phenol red appears red); A milky white opaque precipitation ring surrounds the colony (the strain secretes lecithin enzyme to decompose yolk); Colonies are flat, rough, and have irregular edges, spreading and growing.
